Pharmacokinetics It is absorbed moderately well from the gastro-intestinal tract. It is a potent and highly selective serotonin (5HT) uptake inhibitor . It has no affinity for adrenoreceptors, histamine, GABA 'B' or muscarinic receptors. Indications Fluoxetine is used to treat depression. It is also used to treat obsessive-compulsive disorder, bulimia, narcolepsy and cataplexy. Routes of Administration and Dosage Oral dosage:start with 20 milligrams (mg) a day o.d and can be increased upto 80 mgs/day. Contra Indications It is contraindicated in severe renal failure, lactation,hypersensitivity and concomitant use of MAOI's. Precautions Precautions to be taken in patients with epilepsy, liver failure,cardiac disease, diabetes and in pregnancy. Interactions Anticoagulants, digitalis glycosides : Varying blood levels of these drugs or fluoxetine may occur. Central nervous system (CNS) depressants : The CNS depressant effects may be potentiated. Monoamine oxidase (MAO) inhibitors : Taking fluoxetine while taking or within 2 weeks of taking MAO inhibitors may cause confusion, agitation, restlessness, stomach or intestinal symptoms, sudden hyperpyrexia, severe hypertension, and severe convulsions; at least 14 days should be allowed between stopping treatment with an MAO inhibitor and starting treatment with fluoxetine; if the patient has been taking fluoxetine, at least 5 weeks should be allowed before starting treatment with an MAO inhibitor. Phenytoin : Taking this drug with fluoxetine may result in higher blood levels of phenytoin, which increases the chance of serious side effects. Tryptophan: Taking this drug with fluoxetine may result in increased agitation or restlessness and stomach or intestinal problems.
